Do you have an access to microscopes? Then you can check the size of stomata guard cells. The size of cells are correlated with nucleus size in many plants (but not always). One person in slippertalk mentioned that it is the case in some Paphs.
I've done chromosome counting (not in orchids), but it is super tedious. Flow-cytometry is the easier way to determine ploidy, and that's what most of us uses.
