Welcome to the OB! Glad you are here. Tissue cultures!! Wow, that's pretty cool. I always wondered whether there was a "kitchen" option for meristem cloning....ooo, that's another can o' worms, now isn't it!

Thank you hope we share and discuss a lot of orchid issues, as James wants to see photos of my work and lab, i think i can mange that, maybe after this weekend.
I think you can do Meristem in the "kitchen"why not would there be a problem? i have done it without any contamination at all, but this has to done very quickly (the transplant) simply put three pots of boiling water in a small triangular shape, spray the mother and replating flask with ethanol, remove the ex plant, cut open the the meristem quickly dipping the blade in alcohol every few seconds. it is very difficult to do this all the time, but it can be done well with practice and is good some of like us where equipment, medias, instruments and such like things are rare or never come by.

Wow! Thanks for the info! That's great to know. I have been wanting to experiment with my orchids, but was too afraid that contamination may occur. I guess it is still possible, but sounds like that works pretty well for you. I'll have to give it a try when my orchids permit it. Thanks again.

de-flasking service in Hawaii
I understand there is a good de-flasking and growing service in Hawaii that will take about a year off the growing process. Does anyone know of such a service?
They de-flask your plants and grow them at their location for about a year and then ship the small individual plants to you.

Hi,
I have used old hexangonal or squarish jars, because they rest on their sides without rolling while the agar is setting up. Resting the jar on it's side also prevents any condensation from dripping down on one's developing protocorms.
Just FYI, I deflasked some phal. pulcherrima var. chumpornensis on May 2, 2010...the seedlings are in a compot on a bench in the greenhouse and seem to be thriving! I am thrilled!

i've been hobby flaskin for about 10+ years now and have a big collection of encyclias and caribbean species that are very hard to find in the commercial market.. trying and suceeding to get these out to others so everybody can enjoy them....
becuase once there gone thats it is gone

I mix up Phytagel (an agar substitute) and Phytamax orchid maintenance medium. I mix them, according to directions (kind of like making jello), pour the liquid into the jars. Then, sterlize and set them aside to gel. I have used both an autoclave and a pressure cooker for sterilization purposes and found both to be effective. Are you planning on giving it a try?
