Hi Charles

Start with a sterile box. Use it to propagate succulents
and african violets under sterile conditions... Then move
towards tiny orchid seeds. Be prepared to lose a lot and
on each flask won, be prepared to lose some more.

It's not a Nobel brains matter... it's just a practice. You
know good surgeons started as hair cutters and barbers
and in UK they still do not call FRIS (fellow or the Royal
Institute of Surgeons) doctors but just Mister.

Hi Everyone...I have completed my laminar flow cabinet and just wanted to post a couple pictures and thank you for your help. It cost a little more than I hoped but I know it will be worth it. The filter was about $170 and the fan and switch about $135 and the plastic $60. The rest I had laying around. I have tested it by putting and open flask in it for about 10 min, turning it to catch the air flow and after two weeks, no sign of contamination. I am waiting for an order of seeds to try. Thanks again.

Nice box John. Mine is made from a 58 ct plastic storage container and a computer fan. I can get away with the smaller fan due to the smaller outlets of my box.
I do need to test it though. I can feel the air flowing from the small spaces.
I should probably cook up some test flasks. Funny, I have not done this yet because I have not done this yet. I have a hidden obstacle in my mind about it. I am not sure how I will sterilize my stuff. I do not have an autoclave. I was thinking of using the oven, a large pot or the microwave.
And when mixing agar, does anyone know which to use for which species? What works best for which?
Any suggestions?

Hi Charles Good to hear from you. Can you post a picture of your setup? I would like to see it. Since I am new to this, I have been purchasing media from Phytotech Labs pemixed. They have different formulas depending on the type and species you are trying to sow. Also there is different media depending on whether you are sowing seed or replating your seedlings. They have been very helpful in answering questions about proceedures and such. In general, you can buy premeasured powder to make 1 liter of media. To mix, add the powder to 1 liter of boiling distilled water. I use a wisk to get it mixed well. The media is then placed into clean jars with the lids slightly loose to relieve pressure and into the pressure cooker. Cook at 15lbs steam pressure for 15 min. When cooled you are set to go. I put aluminum foil over the lids before they go into the cooker and I leave it on untill the jars are being opened to sow or receive small plants. I have been using pint jars and I get about 7 jars per liter of replate media. When you get ready to test your flow cabinet, make up the jars of media and let them sit for say a week or two to make sure that the sterilization worked. Then you can open one inside the flow area for 10 min or so, seal it and wait again a week or two to see if there is contamination. There are other points to this that I have learned but I don't want to go on forever. Any questions just ask. There are alot of others on this forum that have more experience then I have and I am sure they will jump in if needed. Either Phytotech's website or the Orchid Seedbank project site has information on sterizing the media in the microwave. I haven't tried it because I like the results from the pressure cooker.

Thanks John. It sometimes helps getting the info from someone within your experience level. The point of view is the same. Same concerns.

I'll try to get some photos as soon as work allows. Don't laugh too hard. I do keep that big wooden club nearby in case of dinosaurs.
