Hi Rody, It depends of the species. With some a little NAA will help in germination. With others it may inhibit germination. Optimal amount? Well, that’s probably in Dr. Arditti’s book too. I’ll leave that question for the experts: -) Just my opinion, I would leave it out for Cattleya unless, others found it to be beneficial.

NAA helps with Vanda. Reference (Mathews and Rao 1980). Helps with Phalaenopsis reference (Baker et al 1987).

Paphiopedilum and Dendrobium NAA is probably not good for germination. Reference (Kano, 1965) and Miyazaki and Nagamatsu (1965). Cleisostoma not good. Reference (Temjensangba adn Chitta Ranjan Deb 2005)

This is very interesting. Thanks Roby for posting!

I also am interesting in tissue culturing aquatic plants. They are very hard to sterilize without killing them. I am going to try sterilizing them this way.

A month ago, I saw 6 flask with Phal tetraspis X Phal violacea on P668 media... following your instruction... so far I got 4 contamination, 2 are germinating.... next time, I will use your method but in a glove box to increase the chance of succes! Thanks! ;-)

Remember that the primary source of contamination is our breath, our hair. The glove box is fine.
