Thanks a lot I will have to try that next time and see if it works mine came out 98% sterile except for the ones that looked bad already and were brown. So what I learned from that is that you should always use fresh stem. Thanks a lot once again!

thanks gix, i've been wanting to get a spawn culture of some type of mushroom for my terrarium. I havn't found what kind would look cool in their yet, but i have tried, and its HARD to get the stuff sterile. That was about 5 years ago so im a little tidier now. I do have all the supplies so i might give that a go here in a few weeks when schools out.

No problem. I do all my sterilizing in the oven, I don't even own a pressure cooker. As soon as it's done baking (and I shut off the oven), I immediately lock the handle on the oven door (like you would do during a self-clean). This keeps any unsterile air from being sucked into the oven during the cooling process. After they cool, quickly open the door and screw (or push) all your lids on tight, and you are good to go. I'll usually make a whole liter of media at once, that way I always have media ready for harvested seeds. I just keep them all labeled in a dark cabinet, ready to go. Good luck to both of you!
