HI ALL MY FRIEND IN THIS FORUM. REALLY I AM A HOBBIST IN ORCHID SEED SOWING AND MERICLONING. I AM DOING MY HOBBY AT HOME, WHAT YOU CALL SOMETHING LIKE" KITCHEN CULTURE " ! IT HAS BEEN REALLY HARD TO STARTWITH ORCHIDS SEED SOWING. MY VERY FIRST QUESTION TO YOU: FOR WHAT PARTICULAR REASON THERE ARE SO MANY VARIATIONS OF THE FORMULA OF THE ORIGINAL MEDIUM OF MURASHIGE AND SKOOG PLANT SCIENCE 1962 ??? IN SOME PARTICULAR SITUATIONS I UNDERSTAND WHY, BUT IN GENERAL THE ORIGINAL FORMULA IS UNIQUE ! MAY I HAVE YOU OPINION ? THANKS IN ADVANCE.

Well first of all welcome to the orchid board from the OCRC in China.
Murashige and Skoog (1962) is today still a good vitro formula; and it has been modified many times. Variations are simply created to improve it or modify it to try and gain maximum results and many times just because of lack of chemicals needed. Knudson formula is a good example; Knudson, Knudson B, Knudson C, Knudson C (Knudson, 1946), Knudson C (Stoutamire, 1964), Knudson C (Morel, 1965), Knudson C (Arditti, 1982), Knudson C (Anderson, 1990) many vitro formulas have been modified for orchid seed sow, replate, tissue. Truly the more they are used and documented (AKA information made public) the better understanding we will have in orchid propagation. We at the OCRC have collected some 145 vitro formulas and we try one seed capsule in many different vitro formulas to see what one can produce the most optimum results. There are many good books you can read about this; I recommend Dr, Arditti books like micrpropagation, orchid biology and review. If you go to Orchid Board social group and you will see Flasking ect…….. is a place that you can ask question and share ideas. Just a short answer to your question. |

MS medium is probably the most common plant tissue culture medium in use for the last 50 years. There are many variations just because it has been used in so many ways for so long. It is commonly seen with or without minor salts, with or without organics, and used in different concentrations, and different sugars or amounts of sugars. Besides that there are probably hundreds of variations that have been used for specific purposes, or sometimes substitutions with alternative salts because of availability or convenience.
