A little background, this flask was sown in Oct 2022, and just reaching 4 months. The protocorms have started on roots and leaves.

Should I replate now? Or wait another month or so?

I am out of my element here, but since no one else has replied, I'd say they look ok to keep going until they are larger and have used up the nutrients in the agar. You will notice that growth slows when they get to that point.

Beauty! I’ve read that replating can be done at a variety of stages all the way from pinhead-sized protocorns to larger protocorns that can be selected for strength/size and all the way to more differentiated plantlets with leaves and roots. Replate medium will have a different balance of nutrients geared to helping the plants produce more leaves and roots. In my limited experience, I would replate those to spread them out and to provide them with the maturation medium to help accelerate their development. What germination medium did you use, btw?

Additionally, you could leave some on the initial plates to see how the progress in comparison…

I just replated a few phal plantlets and protocorns 10 days ago. The plantlets are all now pushing true roots.

This one had a root nub which has quintupled in size whereas on the germ medium it was moving super slow.

Take all that as you will as my experience is limited. Nice to see others working on seed. Would also be nice to have more of a discussion going on the board to exchange ideas and what is working/not working, etc.

Yours are so cute. I think imma wait just another month or 2 before I replate. I def have to mentally prepare cuz this is my first attempt at flasking and I def don’t want to lose it to contamination. Lol

If you’re not using it already, you should get some plant preservative mix (ppm) to add to your media post autoclave. It’s a biocide that will cut your risk of contamination way down. Plant Preservative Mixture (PPM) 30 Milliliters

You’re welcome. You can add it to any media, though I don’t know that you’d want to add it to already solidified medium if that’s what you’re asking. As far as adding it post autoclave, it apparently can’t be sterilized with media containing a certain amount of proteins which both P723 and replate P748 do. I’m guessing it impacts its efficacy. I add after the media has cooled a bit right before I pour. If doing no pours, you would add before they set up. I use an insulin syringes to measure and add as they’re 1ml….the stuff is super strong and used at 1ml/L.

Oh interesting. I’ve never done the pour method, but definitely down to try. Do you do anything to your flasks prior to pouring? And when you pour do you do it out in the open like a kitchen setting, lab, or in a hood?
