You can also do this when the spike is young, before the buds are fully formed. The general point of the matter is, you need to obtain meristematic tissue. It's the immature, undifferentiated cells you're after. Mature, differentiated cells don't provide diddly squat. It doesn't matter where the meristematic tissue comes from.
